G2CMine Data Warehouse

G2CMine integrates the scientific findings of the Genes to Cognition Programme that utilised neuroproteomics, psychiatric genetics, high-throughput mouse gene targeting combined with behavioural and electrophysiological phenotyping and informatics in order to develop a general strategy for understanding cognition at the molecular, cellular and systems neuroscience levels.

G2CMine provides comprehensive Gene Ontology, Mammalian Phenotype Ontology, Human Phenotype Ontology, UniProt, genetic and protein interactions, and regional mouse brain expression results, together with the phenotyping results of the G2C Programme.

Mutation

E14TG2a mouse embryonic stem (ES) cells were targeted with a vector containing 8.7kb and 2.7kb of flanking genomic DNA. This replaced 3.1kb of Gda genomic DNA (X21503036 to X21506136; Ensemble Build 50) with IRES-lacZ-neo cassette. Correctly targeted ES cells were identified by long range PCR using Expand Long Template PCR system (Roche Cat 11681842001). The PCR contained primer X (5’-GAGCTATTCCAGAAGTAGTGAG-3’) and primer Y (5’- CTCAGCTAGAGTGTACTTTAG -3’) that correspond to sequence in the IRES-lacZ-neo cassette and sequence outside the 2.7kb flanking region respectively.

The correctly targeted ES cells were injected into C57BL/6 blastocysts to create chimeric mice, which were bred with 129S5 mice to generate heterozygous (+/–) Gda mutant mice. Those F1 heterozygous mice had been backcrossed with 129S5 mice for 1-2 times before being used for intercrossing.

Location of Gda gene trap. Gda is a 14 exon gene encoding a protein that contains a Guanine deaminase domain (top). We replaced most of Gda exon 3 with a selection cassette in targeted mice and created a frameshift between exons 3 and 4. Primers used for targeted clone identification (x&y) are shown.

Genotyping

Genomic DNA was isolated from ES cells by Wizard SV 96 Genomic DNA purification system (Promega Cat A2371). Genotyping PCR consisted of a 299bp product amplified from the wild-type (wt) allele using a forward primer A (GTCAAGCAAAGACAACCAGAG) in the wt sequence, deleted by targeted mutation, and a reverse primer B (GCTAACTGTCAAGACCTATCC) downstream of the cassette. A 456bp product was amplified from the targeted allele using primer B with forward primer X, within the selection cassette. After enzymatic amplification for 35 cycles (45 seconds at 94 °C, 45 seconds at 55 °C, and 1 minute at 72 °C), the PCR products were size-fractionated on a 2% agarose gel in 1x Tris borate-EDTA buffer.

Primers used for genotyping (a,b & x). PCR genotyping of targeted CYPIN mice using a common reverse primer, b, and forward primers a&x to amplify the wt and mutant alleles respectively.

Expression

Hippocampi were dissected from three wildtype and three heterozygous mutant mice and expression of CYPIN protein was assayed by Western blot (Primary, anti-CYPIN Santa Cruz sc-11699 1:1000; Secondary, anti-goat HRP Abcam ab6885 1:30000; 15min exposure). Expression levels were generally lower in heterozygous mutants (0.166 ± 0.013 s.e.m., p = 0.09, one-tailed T test) than wildtypes (0.200 ± 0.016 s.e.m.). Confirmation on homozygous animals showed significantly lower expression in mutant animals (0.021 ± 0.014 s.e.m., p = 0.0001, one-tailed T test) than in wildtypes (0.232 ± 0.005 s.e.m.).

Breeding

Birth of CYPIN-/- mice followed Mendelian ratios with 20% of offspring being homozygous knockouts. Genotypes of 3-week-old pups from CYPIN+/- intercrosses identified 78 wt, 159 CYPIN+/- and 61 CYPIN-/- progeny (Χ2 p= 0.194). Male and female CYPIN-/- mice showed increased mortality rates between birth and 12 weeks, exhibited small body size but with no gross abnormalities. CYPIN mice were maintained by backcrossing onto the 129S5/SvEvBrd background; heterozygous males and females were fertile and used to set up intercrosses to generate homozygous and wildtype mice to study.

Overview

Mutant mice showed little overall behavioural difference from wildtypes, with no behavioural variables significantly affected by this mutation.
